Signal transduction begins when the peptide binds GHSR-1a on pituitary somatotrophs. The receptor couples to Gq/11 proteins, activating phospholipase C, which cleaves phosphatidylinositol bisphosphate into inositol trisphosphate and diacylglycerol. Inositol trisphosphate releases calcium from intracellular stores, and the resulting rise in cytosolic calcium drives growth hormone vesicle fusion. Concurrent Gs coupling and cyclic AMP elevation have also been reported, and the relative contribution of each arm to the overall secretory response is not fully settled.
Structural features distinguish the molecule from earlier secretagogues. An alpha-aminoisobutyric acid residue near the N-terminus and a D-naphthylalanine substitution increase receptor affinity, while C-terminal amidation improves resistance to exopeptidases. These modifications are associated with reduced stimulation of appetite and of the hypothalamic-pituitary-adrenal axis compared with hexarelin or growth hormone releasing peptide-6. Whether the same profile applies at every dose level studied is a matter of ongoing investigation rather than settled consensus.
Ipamorelin is a synthetic pentapeptide that belongs to the growth hormone secretagogue class. It acts as an agonist at the ghrelin receptor, also called the growth hormone secretagogue receptor type 1a. The compound was designed in the 1990s during a search for agents that release growth hormone with fewer off-target hormonal effects than earlier secretagogues. It appears in the research literature under several sequence-based names. Material supplied for laboratory work is normally a lyophilized solid, and it is not marketed as an approved therapeutic in major jurisdictions.
The molecule contains five residues, including alpha-aminoisobutyric acid, D-2-naphthylalanine, and D-phenylalanine, and it ends in a lysine amide. Non-natural and D-configured residues make the chain less susceptible to common peptidases, which helps explain its resistance to rapid breakdown. Its molecular formula is C38H49N9O5, corresponding to a free-base mass near 711.9 daltons. The C-terminal amide removes a negative charge and is a recurring feature in receptor-active peptides of this family. These structural choices are usually discussed as the basis for its selectivity profile.

At the molecular level, ipamorelin acts as an agonist at the growth hormone secretagogue receptor, also called the ghrelin receptor or GHS-R1a. Binding to this receptor on pituitary somatotroph cells triggers a signaling cascade that leads to growth hormone release. The effect is mediated through phospholipase C and calcium mobilization rather than through the cyclic AMP pathway used by growth hormone releasing hormone. The two pathways are complementary, and combined stimulation produces a larger response than either alone.
Compared with other secretagogues such as GHRP-2, GHRP-6, and hexarelin, ipamorelin is described as more selective. Published animal work reports little or no increase in adrenocorticotropic hormone, cortisol, or prolactin at doses that release growth hormone. This selectivity is the property most often cited in the research literature. Whether the same profile holds across species and dosing schedules remains an open question, since human data are limited and come largely from small studies.
Identity and purity are assessed by complementary methods rather than a single test. Reversed-phase high-performance liquid chromatography separates the peptide from related impurities and reports a percentage purity. Mass spectrometry, most often with electrospray ionization, confirms the expected molecular mass and detects sequence-related variants. Amino acid analysis can verify composition, while water content and residual counterion measurements support the mass balance of a batch. Stability studies under accelerated conditions are used to estimate shelf life, though such estimates carry uncertainty for long-term storage.
Material supplied for research use is normally a white to off-white lyophilized powder. The solid is hygroscopic and is handled in a low-humidity environment to limit water uptake. Bulk quantities are frequently shipped in sealed vials under inert gas. Once reconstituted in water or a neutral buffer, the solution is less stable than the dry powder and is usually divided into single-use aliquots.
At the receptor level, ipamorelin binds GHS-R1a and triggers signaling through Gq-coupled pathways. Activation leads to calcium release and downstream effects in pituitary somatotroph cells. These events promote the release of growth hormone into circulation. The response depends on the presence of the receptor and on the physiological state of the animal or tissue studied. Because the receptor is also found in other tissues, effects beyond the pituitary have been examined in laboratory models, though the extent of those effects remains an area of ongoing study.
One distinguishing feature reported in animal studies is selectivity. Ipamorelin stimulated growth hormone release with limited elevation of adrenocorticotropic hormone or cortisol compared with earlier secretagogues such as GHRP-6. This pattern has been described as more selective for the growth hormone axis. The finding comes mainly from preclinical work, and the degree to which it holds across species and doses is not fully settled. Reports also describe effects on gastric motility in animal models, suggesting activity outside the pituitary, though the clinical relevance of this observation is uncertain.

Relative quantification methods include isotope-coded affinity tags (ICAT), isobaric labeling (tandem mass tags (TMT) and isobaric tags for relative and absolute quantification (iTRAQ)), label-free quantification metal-coded tags (MeCAT), N-terminal labelling, stable isotope labeling with amino acids in cell culture (SILAC), and terminal amine isotopic labeling of substrates (TAILS). A mathematically rigorous approach that integrates peptide intensities and peptide-measurement agreement into confidence intervals for protein ratios has emerged. Absolute quantification is performed using selected reaction monitoring (SRM).
==== Opioid receptors ==== CPA has been found to bind to several of the opioid receptors, including the μ-, δ-, and κ-opioid receptor subtypes. However, this binding is very weak relative to its other actions (IC50 for inhibition of [3H]diprenorphine binding = 1.62 ± 0.33 μM). It has been suggested that activation of opioid receptors might be involved in the sedation that is reportedly sometimes seen with high doses of CPA or in its reported effectiveness in the treatment of cluster headaches.

=== Structural superposition === The most basic possible comparison between protein structures makes no attempt to align the input structures and requires a precalculated alignment as input to determine which of the residues in the sequence are intended to be considered in the RMSD calculation. Structural superposition is commonly used to compare multiple conformations of the same protein (in which case no alignment is necessary, since the sequences are the same) and to evaluate the quality of alignments produced using only sequence information between two or more sequences whose structures are known. This method traditionally uses a simple least-squares fitting algorithm, in which the optimal rotations and translations are found by minimizing the sum of the squared distances among all structures in the superposition. More recently, maximum likelihood and Bayesian methods have greatly increased the accuracy of the estimated rotations, translations, and covariance matrices for the superposition. Algorithms based on multidimensional rotations and modified quaternions have been developed to identify topological relationships between protein structures without the need for a predetermined alignment. Such algorithms have successfully identified canonical folds such as the four-helix bundle. The SuperPose Archived 2015-10-31 at the Wayback Machine method is sufficiently extensible to correct for relative domain rotations and other structural pitfalls.
As (R)-MDMA is less neurotoxic than (S)-MDMA and MDMA or even non-neurotoxic, it may allow for greater clinical viability and prolonged regimens of drug-assisted psychotherapy. (R)-MDMA and (S)-MDMA have shown equivalent effects in terms of inducing prosocial behavior in monkeys. However, (S)-MDMA shows higher potency, whereas (R)-MDMA shows greater maximal effects. Conversely, (S)-MDMA does not increase prosocial behavior in mice, whereas both MDMA and (R)-MDMA do so. MDMA and (S)-MDMA increase locomotor activity, a measure of psychostimulant-like effect, in rodents, whereas (R)-MDMA does not do so. (R)-MDMA likewise showed fewer reinforcing effects than (S)-MDMA in non-human primates. These findings further add to (R)-MDMA showing reduced psychostimulant-like and addictive effects compared to MDMA and (S)-MDMA.
Cinoxacin is a quinolone antibiotic that has been discontinued in the U.K. as well the United States, both as a branded drug or a generic. The marketing authorization of cinoxacin has been suspended throughout the EU. Cinoxacin was an older synthetic antimicrobial related to the quinolone class of antibiotics with activity similar to oxolinic acid and nalidixic acid. It was commonly used thirty years ago to treat urinary tract infections in adults. There are reports that cinoxacin had also been used to treat initial and recurrent urinary tract infections and bacterial prostatitis in dogs. however this veterinary use was never approved by the United States Food and Drug Administration (FDA). In complicated UTI, the older gyrase-inhibitors such as cinoxacin are no longer indicated.
Once the apples are gathered from trees in orchards they are scratted (ground down) into a pulp. Historically this was done using pressing stones with circular troughs, or by a cider mill. Cider mills were traditionally driven by the hand, water-mill, or horse-power. In modern times, they are likely to be powered by electricity. The pulp is then transferred to a cider press where the juice is extracted. Traditionally, the method for squeezing the juice from the apple pulp involves placing sweet straw or haircloths between stacked layers of apple pulp. Today, apples can be quickly pressed on a mechanised belt press. The juice, after being strained in a coarse hair-sieve, is then put into either open vats or closed casks. The remaining solids (pomace) are given to farm animals as winter feed, composted, discarded or used to make liqueurs.

These benefits make nixtamalization a crucial preliminary step for further processing of maize into food products, and the process is employed using both traditional and industrial methods in the production of tortillas and tortilla chips (but not corn chips), tamales, hominy, and many other foodstuffs.
=== Cancer treatment strategies === Tew has studied cancer strategies to devise new treatments. In a joint research, he highlighted the significance of microsomal glutathione transferase 1 (MGST1) in melanin biosynthetic pathways, revealing its role as a determinant of tumor progression, with MGST1 knockdown leading to depigmentation, increased oxidative stress, and hindered tumor growth. He also determined that inhibiting microsomal glutathione S-transferase 1 (MGST1) in melanoma enhances oxidative stress, increases sensitivity to anticancer drugs, and reduces metastasis, improving the effectiveness of therapies. Tew examined ME-344, a second-generation isoflavone with anticancer properties in 2019, demonstrating its impact on redox homeostasis, mitochondrial function, and specific targeting of heme oxygenase 1 (HO-1) in lung cancer cells. In 2020, he determined that ME-344 targets VDAC1 and VDAC2 in lung cancer cells, leading to ROS generation, Bax translocation, cytochrome c release, and apoptosis, highlighting their potential as therapeutic targets. He also assessed how reactive oxygen species (ROS) play a dual role in cancer evolution, influencing both tumorigenesis and cell death, and highlighted tumor cell adaptations in metabolism and antioxidant defenses to manage ROS levels during different stages of cancer development.
=== Pharmacokinetics === The oral bioavailability of amphetamine varies with gastrointestinal pH; it is well absorbed from the gut, and bioavailability is typically 90%. Amphetamine is a weak base with a pKa of 9.9; consequently, when the pH is basic, more of the drug is in its lipid soluble free base form, and more is absorbed through the lipid-rich cell membranes of the gut epithelium. Conversely, an acidic pH means the drug is predominantly in a water-soluble cationic (salt) form, and less is absorbed. Between 16-20% of amphetamine circulating in the bloodstream is bound to plasma proteins. Following absorption, amphetamine readily distributes into most tissues in the body, with high concentrations occurring in cerebrospinal fluid and brain tissue. The half-lives of amphetamine enantiomers differ and vary with urine pH. At normal urine pH, the half-lives of dextroamphetamine and levoamphetamine are 9–11 hours and 11–14 hours, respectively. Highly acidic urine will reduce the enantiomer half-lives to 7 hours; highly alkaline urine will increase the half-lives up to 34 hours. The immediate-release and extended release variants of salts of both isomers reach peak plasma concentrations at 3 hours and 7 hours post-dose respectively. Amphetamine is eliminated via the kidneys, with 30–40% of the drug being excreted unchanged at normal urinary pH. When the urinary pH is basic, amphetamine is in its free base form, so less is excreted.
Ceramic petrography (or ceramic petrology) is a laboratory-based scientific archaeological technique that examines the mineralogical and microstructural composition of ceramics and other inorganic materials under the polarised light microscope in order to interpret aspects of the provenance and technology of artefacts. The process of ceramic petrography involves careful sample preparation. Small sections of the ceramic material are carefully ground down to a thickness of approximately 0.03 mm and then mounted on glass slides. These thin sections allow for the examination of the internal structure of the ceramics and facilitate the identification of mineral phases, crystalline structures, and textural features. The methodology of ceramic petrography draws upon principles from various fields, including optical mineralogy, thin section petrography, and soil micromorphology
Before the formation of Alice in Chains, Layne Staley, a drummer at the time, landed his first gig as a vocalist when he auditioned to sing for a local glam metal band known as Sleze after receiving some encouragement from his stepbrother Ken Elmer. Other members of this group at that time were guitarists Johnny Bacolas and Zoli Semanate, drummer James Bergstrom, and bassist Byron Hansen. This band went through several lineup changes culminating with Nick Pollock as their sole guitarist and Bacolas switching to bass before discussions arose about changing their name to Alice in Chains. This was prompted by a conversation that Bacolas had with Russ Klatt, the lead singer of Slaughter Haus 5, about backstage passes. One of the passes said "Welcome to Wonderland", and they started talking about that being a reference to Alice in Wonderland, until Klatt said, "What about Alice in Chains? Put her in bondage and stuff like that." Bacolas liked the name "Alice in Chains" and brought it up to his bandmates; they agreed and decided to change the band's name. Due to concerns over the reference to female bondage, the group ultimately chose to spell it differently as Alice N' Chains to allay any parental concerns, though Staley's mother Nancy McCallum has said she was still not happy with this name at first. According to Bacolas, the decision to use the apostrophe-N combination in their name had nothing to do with the band Guns N' Roses. The name change happened a year before Guns N' Roses became a household name with their first album, Appetite for Destruction, released in July 1987.

Both molecules activate the same receptor, GHSR-1a, but they share little sequence identity. Ghrelin is a 28-amino-acid hormone carrying a distinctive acyl modification, whereas ipamorelin is a short synthetic peptide. The shared target explains overlapping endocrine effects, while the different structures account for differences in metabolic stability and receptor selectivity.
Early animal and human studies reported growth hormone release at levels that produced small changes in cortisol, prolactin and adrenocorticotropic hormone. That contrast with older secretagogues gave rise to the selectivity label. The observation is consistent across several reports but rests on a limited number of small studies.
Plasma peptidases cleave the molecule into smaller fragments, and reported half-lives in human studies are on the order of a couple of hours. The identity and activity of individual breakdown products have not been fully mapped. Questions about tissue-level clearance remain open.
It has not been approved as a therapeutic by major regulators, and the human trial record is small and dated. Material available today is mostly sold as a research chemical for laboratory use. Approval and restriction status varies by country.
